MAJOR FIELD OF RESEARCH WORK DONE :
- Etiology and management of basal stem rot disease of coconut.
- Etiology, epidemiology, molecular detection and management of coconut leaf rot disease.

Salient findings:
Coconut Root Wilt Disease:
- Standardized the method for the isolation of DNA from coconut tissues (meristem, spindle leaves, inflorescence, petiole and roots).
- PCR conditions were optimized to amplify coconut root wilt pathogen
- DNA from coconut root wilt pathogen, Phytoplasma, was consistently amplified with Phytoplasma specific primers (P4 / P7).
- Cloning and subsequent sequencing of PCR products were carried out using TOPO TA cloning kit (Invitrogen TM).
- Sequenced products were analyzed using BLAST.
- New specific primers were designed
Coconut leaf rot disease:
Studied the phylloplane fungi of leaf rot infected coconut palms and documented the species composition of leaf rot disease (LRD). The population dynamics of the LRD fungi was also investigated and their pathogenicity established. Some of the other major findings on LRD are:
- Colletotrichum gloeosporioides and Exserohilum rostratum were found to be main pathogens based on sequential isolation and seasonal variation in the population of LRD fungi.
- Standardized the technique for evaluating the pathogenicity of LRD fungi in in vitro and in the field.
- Reproduced the LRD symptoms in coconut seedlings by artificial inoculation
- Relative toxicity of some contact and systemic fungicides at different concentrations was assayed in vitro to select an effective broad-spectrum fungicide; Contaf, a systemic fungicide was found to completely suppress the growth of LRD pathogens, Colletotrichum gloeosporioides and Exserohilum rostratum.
- Evolved a simple, more convenient and effective method - pouring the fungicide into the well of spindle - for leaf rot disease control.
- A fast and simple method has been standardized for extracting DNA from LRD infected coconut leaf. A species - specific primer pair for detection of Exserohilum rostratum (Er), erfwd (5' TTA CAC ACA AAA ATA TGA GGG T 3') and errev (5' GAG AGA CAA AAA GAG CCC CA 3') was designed.
- Specific PCR conditions were optimised for the primer pairs, ITS 4 / CgInt and erfwd / errev, for species-specific detection of Colletotrichum gloeosporioides and Exserohilum rostratum, respectively from infected leaf material.
- Using advanced techniques like PCR, RAPD, RFLP and sequencing, variations within the two main pathogens, Colletotrichum gloeosporioides and Exserohilum rostratum were identified to the isolate level.
- Reduction in lesion development was observed on the application of antagonistic bacterium, P. fluorescens with the two main pathogens of LRD, Colletotrichum gloeosporioides and Exserohilum rostratum . The pathogens were applied individually and in combination with the bacterium.
- A total of 118 bacteria (97 from phylloplane and 21 from rhizosphere), 58 fungi (9 from phyloplane and 49 from rhizosphere) and 5 actinomycetes (from phyloplane) were isolated from diseased leaves and rhizotic zones of LRD affected coconut palms.
- All the isolates were subjected to screening for bioefficacy against main fungal pathogens of LRD. Four bacteria and two actinomycetes isolates were found to be effective against pathogens of LRD.
Management of bud rot of coconut:
- Efficacy of the bacterial endophyte Bacillus amyloliquefaciens was proved in vitro.
- The bacterium was found to survive in the leaf axils of coconut for one year. Talc could be used as an effective carrier material for the endophyte Bacillus amyloliquefaciens . Maroti cake supported good growth of the bacterium; thus proving to be an ideal substrate for axil filling.
- Propineb (500ppm) and Ipovalicarp (1000ppm) were found inhibitory to P . palmivora under in vitro conditions.
- PCR procedures were standardized for the detection of P. palmivora with ITS 3 and ITS 4 primers.
- Evolved more simple and effective method - placing mancozeb sachets in the leaf axils (two sachets of 5g each on two sides of the spindle leaf) as prophylactic treatment for leaf rot disease management.
Management of cotton diseases:
- Presently engaged with the programme on the management of cotton diseases using biocontrol agents with special reference to cotton endophytes.
- Attempts are also being made to utilize cotton stalks, which are otherwise wasted by the farmers, for the large scale multiplication of bioagents.
